Laccase (EC 1.10.3.2) is an enzyme belonging to the polyphenol oxidase groups, which plays important role in oxidation of a wide variety aromatic substrates, such as lignin, phenol, polyamine, and aryl diamines, well number other phenolic compounds or inorganic ions presence oxygen. widely applied many different fields, especially textile industry, dyeing, environmental pollution treatment. In this study, we have successfully cloned expressed cDNA coding for laccase from Pleurotus pulmonarius MPN18 (PpLac). corresponds gene (size 1566 bp) was attached pET 21a(+) vector E. coli BL21, after that purified through HisTrapTM sp 5mL column. The PpLac had activity 899.8 U, 74% yield with purity 15.2 -fold, tested by SDS-PAGE electrophoresis molecular weight Mw = 55 kDa. Enzyme displayed optimal at 50 ºC pH 4.0. 20-40 120 min incubation 4 6 h incubation. future, recombinant will be characterized supplementation into cocktail treatment lignocellulosic material.
